Developmental Change of P2Y4 Receptor in Outer Sulcus Cells of the Rat

Abstract
Cochlear outer sulcus cells (OSC) are located between the organ of Corti and K+ secretory cells and known to sustain transepithelial cation currents. It has been thought that P2X2 receptors in OSC provide parasensory K+ efflux pathway in response to intense noise exposure. However, little is known during development. The purpose of this study is to investigate purinergic receptors in developing rat OSC using voltage-sensitive vibrating probe technique. Results showed that there was no response to 100 ¥ìM ADP, UDP, and adenosine at any postnatal age. The response to UTP was greatest at postnatal day 1 (P1) and rapidly decreased to near adult level until P7 when the endolymphatic ion composition is known to be accomplished. This UTP-responsive P2Y receptor was found to be P2Y4, evidenced by the inhibition of UTP effect in the presence of 100 ¥ìM reactive blue 2, an antagonist at rat P2Y4. These temporal changes of purinergic receptors in developing OSC would be associated with cellular differentiation and/or establishment of the endolymphatic ion composition for normal auditory transduction.
